摘要 Polygonum multiflorum Thunb. is widely used in the prescriptions of traditional Chinese medicine (TCM). High-performance liquid chromatography (HPLC) coupled with variable-wavelength detection (VWD) has been used for simultaneous analysis of three anthraquinones [emodin (EM), physcion (PH), emodine-8-O-beta-D-glucoside (EMG)] and one stilbene [2,3,5,4 ''-tetrahydroxystilbene-2-O-beta-D-glucoside (TSG)] in crude P. multiflorum and in the prepared roots (i. e. the preparations Radix Polygoni Multiflori and Radix Polygoni Multiflori Praeparata). Chromatographic separation was performed on a 4.6 mm x 250 mm, 5-mu m particle, C-18 column. The mobile phase was a gradient prepared from acetonitrile and 0.3% (v/v) aqueous formic acid; the flow rate was 1.0 mL min(-1) and the detection wavelength 254 nm. Calibration plots were linear between 3.00 and 150.00 mu g mL(-1) for EM, 1.68 and 84.00 mu g mL(-1) for PH, 1.44 and 72.00 mu g mL(-1) for EMG, and 60.96 and 3,048.00 mu g mL(-1) for TSG, with regression coefficients, r > 0.9992. The intra-day and inter-day precision of the method, as RSD, were less than 1.01 and 2.88%, respectively. Recovery of the four compounds ranged from 101.22 to 102.38%, with RSD less than 2%. This method was successfully used for quantitative analysis of the four target compounds in crude and prepared roots of P. multiflorum Thunb. for quality control.
